* Reactivity of mouse, rat, rabbit, and canine cardiac
troponin-I was demonstrated by dilution of purified
animal cardiac Tn-I (stabilized with rabbit troponin-C)
into serum from the respective species. Experiments
have shown that cardiac Tn-I from mouse and rat give
linear responses to approximately 50 ng/mL. Cardiac
Tn-I from rabbit and dog show linear responses to approx.
20 ng/mL. Please note that the Troponin-I ELISA Kit
is sold for research purposes only and that the kit
was designed for specific measurement of Human Cardiac
Troponin-I.
The Troponin-I ELISA kit uses four monoclonal antibodies
directed against distinct Troponin-I antigenic determinants.
Three antibodies are used for solid phase immobilization
(on microtiter wells). The fourth is conjugated to HRP
and used for detection. The test sample is allowed to
react simultaneously with the four antibodies resulting
in Troponin-I being sandwiched between solid phase and
HRP- linked antibodies. After a 90 minute incubation
period at room temperature the wells are washed with
water. A solution of H2O2/TMB is then added and the
reaction stopped after 20 minutes by addition of 3N
HCl. The absorbance value at 450 nM (or 405 nM) is determined
using a 96-well microtiter plate reader. Human Cardiac
Troponin-I concentration is directly proportional to
A450 (or A405) and is determined by reference to human
cardiac troponin-I standards (0, 2.0, 7.5, 30 and 75
ng/mL) supplied in lyophilized form with the kit.
